Buccal swabs offer the least invasive collection method for clinical or forensic applications. In many cases collection is done using large swabs that require a large volume of wash solution. This is generally done because of the relatively low efficiency of extraction of many DNA purification methods. The ZyGEM technology offers very high efficiency extraction that can typically be done using much smaller swabs, that collect fewer buccal cells but which yield sufficient DNA for hundreds of PCR assays and does not require any concentration steps.
The technology also allows rapid processing of saliva stored on FTA cards with similarly high yields of DNA from the epithelial cells present in saliva. ESR NZ, one of the largest forensic labs in the world has validated and implemented the ZyGEM technology for processing saliva samples stored on FTA paper for use with the highly demanding ABI Indentifiler protocol.
